Modulating Innate Immune Responses for Improved Vaccine Tolerability and Durability

چکیده

Abstract Stimulation of the innate immune system is crucial in both effective vaccinations and immunotherapies. This often achieved through adjuvants, molecules that usually activate pattern recognition receptors (PRRs) stimulate two signaling pathways: nuclear factor kappa-light-chain-enhancer activated B-cells pathway (NF-κB) interferon regulatory factors (IRF). Here, we demonstrate ability to alter improve adjuvant activity viathe addition small molecule “immunomodulators”. By modulating instead receptor binding, these allow customization select responses. We inhibition enhancement products NF-κB IRF pathways by several orders magnitude. Some modulators apply generally across many receptors, while others focus specifically on individual receptors. Modulators boost correlates a protective responses commercial flu vaccine model reduced reactogenicity typhoid model. These have range applications: from adjuvanticity prophylactics immunotherapy. CTI special feature on innate immune responses and vaccine design

T he challenge of developing next generation vaccines against some of the most burdensome diseases—both infectious and inflammatory, will require an injection of basic knowledge as well as scientific innovation. As highlighted in this collection of expert reviews, the innate immune response is a critical stepping-stone between vaccination and the development of effective adaptive immunity, trig...

Local innate immune responses in the vaccine adjuvant-injected muscle

Inducing a high magnitude of antibodies, possibly in combination with T-cell responses that offer epitope breadth over prolonged periods of time is likely a prerequisite for effective vaccines against severe diseases such as HIV-1 infection, malaria and tuberculosis. A much better understanding of the innate immune mechanisms that are critical for inducing desired responses to vaccination would...

Killer Cell Immunoglobulin-Like Receptors Influence the Innate and Adaptive Immune Responses

Natural killer (NK) cells are a subset of lymphocytes which play a crucial role in early innate immune response against infection and tumor transformation. Furthermore, they secrete interferon-γ (IFN-γ) and tumor necrosis factor (TNF) prompting adaptive immu-nity. NK cells distinguish the unhealthy cells from the healthy ones through an array of cell-surface receptors. Human NK cells use inhibi...

HCV Innate Immune Responses

Hepatitis C virus (HCV) establishes a persistent infection in more than 70% of infected individuals. This striking ability to evade the powerful innate immune system results from viral interference occurring at several levels of the interferon (IFN) system. There is strong evidence from cell culture experiments that HCV can inhibit the induction of IFNβ by cleaving important proteins in the vir...

Macrophage PTEN regulates expression and secretion of arginase I modulating innate and adaptive immune responses.

The activation of innate immune cells triggers numerous intracellular signaling pathways, which require tight control to mount an adequate immune response. The PI3K signaling pathway is intricately involved in innate immunity, and its activation dampens the expression and release of proinflammatory cytokines in myeloid cells. These signaling processes are strictly regulated by the PI3K antagoni...
